I am planning to go to lemurs parc. It looks like it will cost a lot to get there by taxi so wondered about turning it I to a full day trip. Is there anything worth combining it with ad a full day out of Tana? I have been quoted 40 USD to take me, wait and bring me back. Says leaving at 8 and returning at 5 but all reviews don't seem to think it needs that long. Any advice appreciated.

You do rent the car for the full day so smart figuring out options. I didn’t know that until too late to plan like you. I left Tana at 9 and was back by 1; spent 1.5 hours at park. Unfortunately i believe the other outside of Tana sights are the opposite direction of the park. You may need to do some google mapping to see about combining. Traffic is bad so it may be difficult. As other responder said, Best to do tourist walk to see the sites in Tana.
Will we need to reserve tickets for entrance or guides in advance?

No, I don't think so. Just show up. Might be busier on weekends but I think there's always a guide. We went in the morning, so maybe that's another consider.
Lemur parc is just outside Antananarivo, the capital. Lemur island is a little reserve owned by Vakona Hotel in Andasibe. That is a 3 hour drive from the capital.
